Estradiol (E2) is produced in women mainly in the ovary. In men, the testes and adrenal glands are the principal source of Estradiol. In women, normal levels of Estradiol provide for proper ovulation, conception, and pregnancy, in addition to promoting healthy bone structure and regulating cholesterol levels. Estradiol levels are used to help evaluate ovarian function. Estradiol helps diagnose the cause of precocious puberty in girls and gynecomastia in men. Its main use has been in the differential diagnosis of amenorrhea (for example, to determine whether the cause is menopause, pregnancy, or a medical problem). In assisted reproductive technology (ART), serial measurements are used to monitor follicle development in the ovary in the days prior to in-vitro fertilization. Estradiol is also sometimes used to monitor menopausal hormone replacement therapy. |

hCG is responsible for maintaining the function of the ovarian corpus luteum. In normal pregnancy, hCG concentrations rapidly increase with a doubling rate of approximately two days. Pathologically decreased values indicate abnormal pregnancy (e.g. ectopic pregnancy). A sudden fall of hCG levels before the 8th–10th week of gestation should be considered as a serious indicator of imminent abortion. If pregnancy can be excluded, hCG levels above normal are highly likely to indicate the presence of malignant neoplasm, particularly germ-cell tumors (e.g. testicular tumors, chorionic or ovarian carcinoma). For use as a tumor marker, the test system has to recognize both hCG and βhCG, since testicular and chorionic carcinomas
mainly secrete the free β-chains rather than the intact hCG molecule. |

Progesterone concentrations are useful in the documentation and timing of ovulation. Progesterone may also be useful in the evaluation of patients with sterility due to luteal phase defects. In order for Progesterone measurement to be useful as a sign of ovulation, it must be obtained as -closely as possible the mid-luteal phase of the cycle.
